Need more info to answer that. The answer depends on what your goal is; i.e. drag truck, street truck, etc. Also, whats your budget. You can do just motor, but if you want to go turbo, you still need a built motor plus the turbo system.
More info!!!

Lots of things you could do.
1. Cam, heads, exhaust, stall, tune.
2. 6.0 with above.
add nitrous to either of above.
3. keep 5.3, do stall, cam, STS, tune, intercooler.
4. forged 6.0, stall, cam, STS or custom front mount turbo with intercooler, tune, rear suspension.

Oh yea, a fuel system with any forced induction.

I have a 6.0L with an STS turbo. This truck is made to be a daily driver and a street truck. Allen set it up for me and the truck runs great. It is only running 5-6PSI, but is capable of running 10PSI with a minor adjustment. If you want a daily driver and a street truck, I would go with an STS turbo intercooled, Nelson tune fuel pump larger injectors, and rear suspension. You will be amazed.
I am doing a stall and maybe a head cam swap soon as well as rear suspension.

My answer is and always will be TURBO! Once you start, though, you will be addicted to boost for the rest of your life, so BEWARE!! By the way me and my bro saw a lightning out crusing the other night and followed him home. he was too scared by the way my truck sounded to even go race.....pussy.
