I would like to put a 5 speed behind my 6.0L, but i'm afraid the NV3500 won't hold up. Anyone know anything about the NV4500? I know Dodge used it for a while

NV4500 is what GM used behind the 6.0

the 3500 will hold. i've seen a truck that's took the v6 out & built a 454 in it's place, beat on it, and then swapped the 454 for an 8.1 and ragged on it some more.
i think the measly torque from your little 6.0 will be safe.

The NV4500 is a great transmission, but it is geared for a work truck, not something you want to be fast.
