I would leave the radiator inline (go from trans to radiator to new cooler, back to trans)
But if you had a factory tow package with the one infront of the radiator, I would just disconnect that and install new one.

blown transmissions are about as useful as 97% of the guys on this forum
the line coming out of the top of the radiator is the return line to the transmission, hook it into it. Run that hose to the bottom inlet of the new cooler. The whole idea is that the fluid has to completely fill the cooler before returning to the transmission.
